Advancement in somatic hybridization in the Potato Research Institute Havlíčkův Brod during the past more than 15 years (A)

Somatic hybridization has been used in potato for 40 years because it overcomes the sexual barriers between the cultivated and wild Solanum species. In this paper, we report on advancement in somatic hybridization in the Potato Research Institute Havlíčkův Brod. To date 168 inter-specific, 25 intra-specific and 12 potential asymmetric somatic hybrids have been produced via protoplast electrofusion of 7 Solanum species. Somatic hybrids have been characterised for a number properties such as ploidy level, cytoplasm type, agronomic characteristic or disease resistance. Several fusion products were exploited in potato breeding.

Potato collection maintained in the in vitro gene bank and its genetic potential

The mission of the potato genetic resource collection of Solanum genus in the Potato Research Institute Havlíčkův Brod is assembly, documentation, long-term conservation and guarantee for potato genetic resources in the Czech Republic. A part of this involves systematic study, especially of new obtained materials, their description and evaluation with recommendation of suitable donors of economically valuable traits and characteristics, development of the national potato database with passport and descriptive data unification based on GRINCZECH system and providing of genetic resources to inland and foreign users. The potato collection involves 2,581 in vitro entries. The collected wide biodiversity of Solanum tuberosum ssp. tuberosum varieties, S. tuberosum tetraploid hybrids and dihaploids combined with conserved accessions of cultivated and wild species represents a wide option for selection of donors possessing important economic and breeding properties for breeders and researchers.

The study describes the technology of virus–free production of breeding and basic propagation material of potato, into that preventive multistep checking of source materials for presence of potato ring rot caused by the quarantine bacterium Clavibacter michiganensis subsp. sepedonicus (Cms) has been recently included. Specific biotechnological techniques are mentioned used for establishment and maintenance of in vitro virus–free collections. The system of rigorous check for potato ring rot prior to in vitro culture establishment enables breaking of vertical disease spreading chain in breeding and basic propagation material of potato.

The identification of PVY isolates by means immunocapture RT-PCR probing

Twenty PVY isolates were characterised. Biologically they were characterised as PVYO (three isolates) and PVYN (17 isolates). However, serological and molecular genetic methods led to a more complicated characterisation. For example, five isolates induced necrotic symptoms on tobacco plants typical of PVYN, despite reacting as PVYO serologically. Moreover, the PVY isolates were not identical according to molecular genetic procedures. Typical PVYNTN PCR products were observed for 14 isolates, but five of them (Hr220-5, Hr387-7, Nord 242, Syn1Scot and 41-97) did not produce potato tuber necrotic symptoms in infected cultivars.
The method of immunocapture RT-PCR probing was developed using a set of 24 pairs of primers derived from eight regions of the PVY genome. Using this method, five out of seven PVYNTN isolates, including the Czech standard PVYNTN from the potato cv. Nicola, were found to be identical. However, two PVYNTN isolates and all the other probed PVY samples showed unique patterns, suggesting specific differences at the nucleotide level. This method enabled specific identification of individual isolates variability even within different PVY sub-groups.

The use of molecular genetic techniques at VÚB Havlíčkův Brod in the area of potato genotyping has begun 20 years ago. At present, these techniques are predominantly used to characterize the hybridity of materials and to monitor the presence of selected genes of resistance in individual genotypes. The aim of this publication is to show developments in this area.

The selected wild Solanum species were chosen for crossing by means of traditional breeding methods. These wild species, maintained in genebank of Potato Research Institute in Havlíčkův Brod, are a good source of resistance to various biotic and abiotic factors. Hundreds of hybrids from many combinations, which were arisen by reciprocal crossing procedures (wild species x variety or dihaploid) and parental genotypes were tested by means of PCR methods. We obtained the crossing materials with wild species (S. acaule, S. bert­haultii and S. demissum) with potato varieties. Hybridity of progeny after interspecific hybridization was visually checked by morphological traits and RAPD method. The hybridity was found at some hybrids from crossing combinations only. By means of PCR genotypes from seven wild species (S. acaule, S. berthaultii, S. bulbocastanum, S. brevidens, S. demissum, S. pinnatisectum and S. verrucosum) were tested. The plant ma­terial was transferred from in vitro conditions to the greenhouse and grown there. The DNA was isolated from these plants by means of GenElute Plant Genomic DNA Miniprep Kit (Sigma). DNA markers were chosen and tested. The selected markers were used for subsequent testing of diploid and tetraploid potato collections. The wild species bearing resistance markers are used for crossing experiments.

Practical experience with potato drip irrigation in the early potato production region between 2017 and 2018

The paper describes the evaluation of two-year trials with drip irrigation at potato varieties Monika and Jolana under conditions of Southern Moravia. Compared to non-irrigated control, a relatively significant yield increase (two-fold to three-fold) was recorded. Drip irrigation is a modern tool for irrigation of potatoes that require balanced water regime for the whole growing season. The variety Monika responded better to the irrigation in both years, reaching higher yields also in non-irrigated variant compared to Jolana.
